      为了解大麦品种 HvPDIL5-1 HvEIF4E 基因的单倍型及其与大麦黄花叶病抗性的关系,以扬农啤系列、扬饲麦系列和其他来源的29个品种(系)为材料,在江苏扬州和盐城病圃进行大麦黄花叶病大田自然抗性鉴定;利用大麦 HvPDIL5-1 HvEIF4E 基因全CDS区的3对引物对供试品种(系)的 HvPDIL5-1 HvEIF4E 基因进行扩增、测序,分析两个黄花叶病抗性基因的单倍型及其抗性。结果表明,本试验年度两试点大麦黄花叶病发病程度较正常年份偏轻,29个品种(系)中有20个品种(系)在扬州和盐城对大麦黄花叶病同时表现为免疫或高抗,Ea52和苏B1403在扬州表现为免疫、在盐城表现为中抗,其余7个品种(系)在两试点均表现为中抗或中感。供试材料的 HvPDIL5-1 基因存在4种单倍型, HvEIF4E 基因存在14种单倍型。扬农啤2号、扬饲麦3号、扬农啤7号等10个品种(系)在扬州和盐城病圃对大麦黄花叶病均表现为免疫或高抗,但其 HvPDIL5-1 HvEIF4E 基因均为BaMMV-ASL感病单倍型,说明这些品种(系)携带有除 rym1/11 rym4/5 以外的其他黄花叶病抗性基因。携带抗性基因 rym5 的苏B1403在盐城病圃鉴定中表现为中抗,说明江苏大麦产区出现了拮抗 rym5 抗性基因的病毒株系,需进一步加强大麦黄花叶病抗性新基因的发掘。

      In order to understand the relationship between the haplotypes of barley genes HvPDIL5-1 and HvEIF4E and barley yellow mosaic disease resistance,in this study,29 varieties(lines) including Yangnongpi series,Yangsimai series and other sources,were used to identify the natural resistance to barley yellow mosaic disease in Yangzhou and Yancheng disease nurseries of Jiangsu Province.The HvPDIL5-1 and HvEIF4E genes of the tested varieties were amplified and sequenced using the primers of the whole CDS region of the HvPDIL5-1 gene and HvEIF4E gene,and the haplotypes of these two yellow mosaic disease resistance genes were analyzed. The results of resistance identification showed that the incidence of barley yellow mosaic disease in 2020 was lower than that in normal years. Among the test materials,20 varieties(lines) showed immunity or highly resistance in Yangzhou and Yancheng; Ea52 and Su B1403 were immune in Yangzhou and moderately resistant in Yancheng,and the remaining seven varieties were moderately resistant or moderately susceptible to barley yellow mosaic disease. The test materials contain 4 haplotypes for HvPDIL5-1 gene and 14 haplotypes for HvEIF4E gene.10 varieties(lines) such as Yangnong 2,Yangsimai 3 and Yangnong 7 showed immunity or highly resistance to barley yellow mosaic disease in Yangzhou and Yancheng,but they all showed BaMMV-ASL susceptible haplotype in HvPDIL5-1 and HvEIF4E ,indicating that these varieties (lines) may carry other yellow mosaic disease resistance genes except for rym1/11 and rym4/5. Su B1403 carrying the resistance gene rym5 in Yancheng disease nursery showed morderately resistanity,indicating that there may contain an antagonistic rym5 resistance gene virus strain in Jiangsu barley producing areas,and the excavation of new barley yellow mosaic disease resistance gene needs to be further strengthened.
