为探究由小麦矮腥黑粉菌基因g4418编码的蛋白的生物学功能,根据小麦矮腥黑粉病菌转录组测序结果,筛选出由基因g4418编码的蛋白,通过提取小麦矮腥黑粉菌总RNA并合成cDNA,然后以cDNA为模板利用PCR技术扩增出g4418目的基因,对其编码蛋白进行了生物信息学分析,通过构建pGR107-g4418融合载体和pBin-GFP-g4418表达载体进行毒性验证和亚细胞定位。结果表明, g4418基因全长432 bp,共编码143个氨基酸;烟草毒性验证表明,该蛋白无毒性;共聚焦显微镜观察亚细胞定位结果显示,该效应蛋白g4418可定位在细胞膜和细胞核。 |

In order to explore the biological function of the effector protein encoded by g4418 gene, the effector protein encoded by g4418 gene was screened according to the transcriptome sequencing results of Tilletia controversa Kühn. The total RNA of Tilletia controversa Kühn was extracted and the cDNA was synthesized. The target fragment of g4418 gene was amplified by PCR using cDNA as template, and the encoded protein was analyzed by bioinformatics. The pGR107-g4418 fusion vector and pBin-GFP-g4418 expression vector were constructed for toxicity verification and subcellular localization. The results showed that the full length of g4418 gene was 432 bp, encoding 143 amino acids. Tobacco toxicity verification showed that the effector protein was non-toxic. The subcellular localization of the effector protein g4418 was observed by confocal microscopy. The results showed that the effector protein g4418 was localized in the cell membrane and nucleus. |
