      miR164是植物中一种重要的非编码RNA,对植物的生长发育具有重要作用。为了解miR164靶基因与小麦衰老的关系,本研究通过在线软件对小麦中miR164家族成员进行了鉴定并对其进行生物信息学和表达分析。结果表明,从小麦中共鉴定出13个miR164家族成员,其不均匀分布在9条染色体上。系统进化分析表明,13个小麦miR164与同属于禾本科单子叶植物的水稻亲缘关系较近。13个Tae-miR164均能形成稳定的二级结构,成熟序列碱基具有高保守性,仅有3个Tae-miR164成员在第21位存在差异,成熟序列产生于前体中具有碱基高保守性的第1~21位。Tae-miR164的靶基因中,有18个为NAC转录因子,占所有靶基因总数的75%,表明Tae-miR164的主要靶基因为NAC转录因子。Tae-miR164家族成员在小麦叶片中的表达量明显高于其他组织;在小麦叶片衰老过程中,与其他Tae-miR164靶基因相比,有3个Tae-miR164靶基因表达差异显著,其ID分别为TraesCS4A02G225100、TraesCS5B02G054800和TraesCS5A02G04910,其中TraesCS5A02G04910为NAC转录因子,推测NAC转录因子与小麦叶片衰老密切相关。比较不同物种中miR164家族成员数量,发现miR164家族成员数量与物种基因组大小和物种亲缘关系无关。
英文摘要:
      miR164 is an important non-coding RNA in plants and plays an important role in plant growth and development. In order to understand the relationship between miR164 target genes and wheat senescence, this study identified members of the miR164 family in wheat through online software and conducted bioinformatics and expression analyses. The results showed that a total of 13 members of the miR164 family were identified from wheat, which were unevenly distributed on 9 chromosomes. Phylogenetic analysis showed that the 13 wheat miR164 were closely related to rice, which belong to monocotyledonous plants of Poaceae. All 13 Tae-miR164 members can form stable secondary structures, and the mature sequence bases are highly conserved. Only 3 Tae-miR164 members have differences at the 21st position, and the mature sequence is generated from the 1st to 21st positions with high base conservation in the precursor. Among the target genes of Tae-miR164, 18 were NAC transcription factors, accounting for 75 % of the total number of target genes, indicating that the main target genes of Tae-miR164 were NAC transcription factors. The expression level of Tae-miR164 family members in wheat leaves is significantly higher than that in other tissues. During the senescence process of wheat leaves, three Tae-miR164 target genes were significantly differentially expressed compared to other Tae-miR164 target genes: TraesCS4A02G225100, TraesCS5B02G054800, and TraesCS5A02G04910. Among them, TraesCS5A02G04910 is a NAC transcription factor, suggesting a close correlation between NAC transcription factors and wheat leaf senescence. By comparing the number of members of the miR164 family in different species, it was found that the number of miR164 family members is not related to the size of the species genome and the distance of species phylogenetic relationship.
