PHO gene family is mainly involved in phosphate transport and plays important roles in plant growth and development. In order to study the function of PHO gene family members in wheat,PHO genes were identified from the whole wheat genome,and their gene structure,conserved domain of proteins,phylogenic relationship,cis-acting elements and expression characteristics were analyzed using bioinformatics methods. The results showed that 12 members of the PHO gene family were screened and identified in the wheat genome database,and all members contained SPX and EXS domains. Through phylogenetic analysis,conserved domains and gene structure analysis,it was found that the PHO proteins of wheat were closely related to PHO1-H1 protein of Arabidopsis and PHO1 protein of rice. Except for TaPHO7,TaPHO10,TaPHO11 and TaPHO12 proteins lacking partial motifs,other PHO proteins have complete motifs,and in the same subgroup,PHO gene family members have similar conserved domains and gene structure,demonstrating that the PHO subfamily members are highly conserved. Cis-acting element analysis showed that the promoter region of wheat PHO genes have hormone induction,light response and low temperature response elements related to phosphorus regulation. Tissue-specific analysis found that most genes were highly expressed in roots based on RNA_seq data. Twelve PHO genes were analyzed by qRT-PCR,and it was found that TaPHO7 and TaPHO8 had higher expression levels in the underground part(root) of Xiaoyan 54 after low phosphorus stress treatment,compared to control. |
