In order to analyze the effect of light on the regulation of vernalization and its mechanism, the flowering time of a wheat cultivar Zhongmai 175 after vernalization in light or dark for 28 d were compared. The leaf samples were also collected during vernalization in light or dark for 0 d, 7 d, 14 d, 21 d and 28 d, and real-time PCR analysis of vernalization- and photoperiod-related genes was performed using these samples. The result showed that compared to dark vernalization, vernalization with light brought anthesis forward by 19.73 days in wheat. The real-time PCR revealed an elevated expression for most of the vernalization and photoperiod related genes(TaVRN1,TaVRN2,TaVER2, TaVIL1, TaVIL2, TaVIL3, TaCO1, TaCO2, TaPPD1, TaGI1, TaLHY and TaTOC1) analyzed in this study under vernalization in light. This may be one of the major reasons for early flowering under vernalization in light in wheat. These results also suggest some hitherto unknown regulatory factors relate to TaVRN1,regulating both by light and low temperature. |
