To investigate the effect of exogenous MeJA on the expression of TaICE41 and four cold-responsive genes(Wcor14, Wcor15, Wcor18, Wcor413) in wheat Dn1 leaves and tillers under low temperature stress, and to reveal whether the regulation of cold response genes in JA to CBF pathway are responsible to the strong cold tolerance, Dn1 was used as the experimental material, and MeJA(1 mmol·L-1) was applied.The expression of TaICE41 and four cold-responsive genes was detected by RT- PCR method in Dn1 leaves and tillers under 5 ℃, 0 ℃, -10 ℃ and -25 ℃.The results showed that exogenous MeJA increased the expression of the above genes in Dn1 leaves and tillers under low temperature. At -10 ℃, the expression levels of TaICE41, Wcor14, Wcor15, Wcor18 and Wcor413 in Dn1 reached to the highest value. The expression of the five genes in the treatment group was 3.3 times, 38.0 times, 12.7 times, 9.4 times, and 3.5 times of that in the control group, respectively, indicating that the regulation of JA on the cold response gene in CBF pathway is related to the strong cold tolerance of Dn1 under low temperature stress. -10 ℃ is an effective temperature for cold stress, and the tillering node is the main part related to wintering.Bioinformatics analysis and prediction showed that TaICE41 is located on chromosome 3A in the nucleus, encoding a random coiled protein which belongs to the HLH protein family.It is closely related to the ICE1 protein of Aegilopstauschii.This study provided a theoretical basis for JA in regulating the cold tolerance of plants. |
