Wheat Fusarium head blight(FHB) is a destructive disease worldwide that threats food safety. The elite germplasms with FHB-resistance are the basis for breeding Fusarium resistant cultivars. In this study,the progenies of 979-5(medium susceptible to FHB) and Sumai 3(high resistance to FHB) were used as materials. By using the method of pedigree breeding,the selection of agronomic characters,SSR markers and the single flower inoculation and identification of FHB,the new FHB resistant germplasm was selected. Results showed that:among the 13 SSR markers closely linked with the major resistant gene Fhb1,and two minor resistant genes Fhb2 and Fhb5,six and seven of the markers show monomorphic and polymorphic fragments when amplified from 979-5 and Sumai 3,respectively,which indicated that 979-5 and Sumai 3 had high genetic similarity at the three loci. Twenty lines carrying Fhb1 were created and selected,among which 13 lines performed medium resistance and seven lines with either Fhb2 or Fhb5 performed high resistance to FHB. The 20 new lines performed not only cold resistance in winter,but also lower plant height,similar growth period with their parent 979-5,good yield traits,and better grain quality characters,such as protein content,flour yield,wet gluten content and dough stability time. Among them,the four quality indices of three new lines:k15,k32 and k40,reached the national standard for medium and strong gluten varieties. The new lines can be used as excellent breeding materials for improving FHB resistant varieties in Huanghuai winter wheat region. |
