      SBP家族是植物特有的一类转录因子,含有一个约80个氨基酸残基的保守结构域,广泛参与植物生长、发育及多种生理生化过程。为探讨大麦SBP转录因子的基因结构及表达模式,采用生物信息学方法对大麦SBP家族的基因结构、染色体分布、蛋白质保守结构域、系统进化树及表达谱进行分析。结果发现,用生物信息学方法鉴定共获得22个大麦SBP基因,命名为 HvSBP1~ HvSBP22,分布在6条染色体上,而且大多基因成簇分布;各基因的结构差异明显,外显子数量为1~11个。蛋白结构分析显示,大多数HvSBP具有2个典型的锌指结构域以及核定位信号。系统进化分析表明,22个HvSBP分属于4个进化亚组,且与水稻SBP基因的亲缘关系较近。表达分析表明,22个HvSBP在萌动胚、幼苗、5 dpa颖果、15 dpa颖果、0.5 cm幼穗花序、1 cm幼穗花序和节间等7个组织中的表达差异明显,呈现出不同的表达模式。研究发现,HvSBP基因主要集中在幼穗花序和颖果等组织中表达,在1.0 cm幼穗花序中表达量最高,表明HvSBP基因与大麦开花发育密切相关。
英文摘要:
      Squamosa promoter binding protein(SBP),as a plant-specific transcription factor,containing an about 80 amino acid conserved domain,plays important regulating roles in the processes of plant growth and development,as well as many physiological and biochemical processes. To explore the function of the barley SBP genes,their gene structure,chromosome distribution,conserved domain of proteins and so on,were analyzed using bioinformatics methods. The results showed that 22 HvSBP genes,named HvSBP1-HvSBP22,were distributed on 6 chromosomes of barley in cluster. The structure of each gene were significantly different,and the number of gene exons varied from 1 to 11. Protein structure analysis revealed that most of HvSBP had 2 typical conserved zinc-binding sites and a nuclear localization signal. Phylogenetic analysis showed that 22 HvSBPs belonged to 4 subgroups,which shared close relationship with those in rice. Expression pattern analysis revealed that 22 HvSBPs expressed differently in seven tissues including germinating embryo,shoot,caryopsis (5 dpa) ,caryopsis(15 dpa),inflorescence(0.5 cm),inflorescence(1 cm) and internode.The study showed that the expression of these HvSBP genes were focused in inflorescence and caryopsis,and they had the highest expression level in barley inflorescence(1 cm). These results suggested that these HvSBP genes were closely related to the flowering and development of barley.
