Psathyrostachys huashanica(2n=2x=14,NsNs) harbors numerous excellent gene resources and exhibits extensive application prospects as a breeding resource.In this research,H1133,H4122 and H1423,derived from common wheat 7182 and P.huashanica,were characterized by cytological analysis,genomic in situ hybridization(GISH),fluorescence in situ hybridization(FISH) and molecular markers analysis.Results showed the chromosome number and configuration of them were 2n=44=22Ⅱ.GISH-FISH analyses indicated they had 42 chromosomes from common wheat,and two Ns chromosomes from P.huashanica.What's more,EST and PLUG marker analyses revealed that H1133,H4122 and H1423 contained 3Ns,5Ns and 7Ns chromosomes of P.huashanica,respectively.Thus,H1133,H4122 and H1423 are wheat- P.huashanica 3Ns,5Ns and 7Ns alien addition lines,respectively.The three lines exhibited different agronomic characters from 7182 in plant height,tiller length and spike length.H4122 and H1423 show stripe rust resistance to the mixture of CYR32 and CYR33 at adult stage,and the resistance gene may originate from P.huashanica according to the pedigree.These mean that H1133,H4122 and H1423 could be used as potentially bridge materials in wheat breeding with specific agronomic traits and disease resistance to stripe rust. |
