      为了有效利用含二倍体长穗偃麦草7E染色体的种质材料,利用荧光原位杂交和分子标记技术以及赤霉病抗性评价,对从中国春-二倍体长穗偃麦草7E代换系DS7E(7B)与扬麦16杂交的F2种子辐射后代中选育的小麦-长穗偃麦草易位系TW-7EL2进行了鉴定。荧光原位杂交(FISH)结果表明,易位染色体的小麦染色体片段与小麦7B短臂的杂交信号相似;分子标记检测结果表明,在易位系中能够扩增出7EL特异条带,但缺失小麦7BL特异条带。综合上述结果,将该易位系命名为T7BS·7EL。且其赤霉病抗性显著高于中国春和扬麦16,与苏麦3号接近,是小麦赤霉病抗病育种的新种质。继续利用1 200 rad 60Co-γ射线辐射处理易位系T7BS·7EL的成熟花粉并授予扬麦158,从其纯合易位系的辐射后代M1中检测到长穗偃麦草7EL染色体的小片段中间插入易位、顶端易位和7EL染色体缺失等结构变异的单株15株,诱变频率为15.62%,表明利用小麦-长穗偃麦草整臂易位系进行成熟花粉辐射,可以高效诱导产生小片段易位材料。
英文摘要:
      In order to effectively utilize the germplasm material with diploid Thinopyrum elongatum chromosome 7E,the translocation line TW-7EL2 was identified from the plant progenies of irradiated F2 seeds from the cross between Yangmai 16 and Chinese Spring-Th.elongatum substitution line DS7E(7B) through chromosome FISH,molecular marker analysis and FHB resistant test. The FISH analysis showed that the hybridization signals of wheat chromosome segment of the translocation chromosome in TW-7EL2 was similar to that of wheat chromosome 7BS.The marker Xgwm333 located on chromosome 7BL amplified Th.elongatum chromosome 7EL specific band in TW-7EL2 and DS7E(7B),but the specific band of wheat chromosome 7BL was absent. The results of FISH and molecular marker analysis showed the translocation line was designated as T7BS·7EL.The resistance level of the translocation line was higher than that of Chinese Spring and Yangmai 16 and similar to that of the resistant check Sumai 3 after a successive three-year field FHB resistant test. The result showed the translocation line T7BS·7EL with the stable FHB resistance and feasibility can be used as a new germplasm for improving FHB resistance in wheat. On the basis of the above studies,the mature pollen of homozygous translocation line T7BS·7EL were irradiated with 1 200 rad 60Co-γ ray before flowering,and then pollinated to common wheat variety Yangmai 158. Among the 96 M1 plants,15 chromosome aberration plants involving 7EL were screened by GISH,including interstitial translocation chromosomes,terminal translocation chromosomes and 7EL deletion chromosomes,and the mutation frequency was 15.62%. These results showed that wheat-Th.elongatum whole arm translocation line of mature pollen irradiation could efficiently induce small segment translocations.
