In order to know the genetic diversity of wheat starch granule associated proteins (SGAPs) in Sichuan wheat, 66 wheat landraces, 100 new wheat cultivars (lines) and 21 wheat founder parents from Sichuan were analyzed by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS PAGE). A total of seven combinations and ten different protein bands were identified in the 187 materials, and the variations, including unexpressed and the change of mobility, were observed on five bands. The null SGP 1A, the null Wx 1B, and the null Wx 1D were detected in 114, 18, and 8 materials, respectively, but the null Wx 1A was only observed in one material. Based on the comparison with SGAPs in Chinese Spring nullisomic tetrasomic lines (N7AT7B, N7DT7B, N4AT4B), and T. urartu(AA), Ae. tauschii(DD), T. turgidum(AABB), and the results of MALDI TOF MS, we further confirmed that the identification of SGAPs was correct. Our result suggested that the new varieties (lines) showed higher diversity on SGAPs than the landraces of Sichuan, and the materials with the variation of SGAPs could be used in the functional research of SGAPs and the wheat breeding on starch quality. |
