      为了明确大麦成株叶锈病抗性品种‘Pompadour’抗性基因的性质,以‘WABAR2482×Pompa dour’的457个F2后代为材料,采用单链构象多态性(SSCP)分析技术,对发掘的SNP EST 分子标记在抗性品种的F2后代中进行分子标记检测。结果表明,457株F2分离群体中328株表现抗病,129株表现感病,卡方检验符合3∶1的分离比例。推断新发现的5HS成株叶锈病抗性基因为主效显性抗性基因。在所筛选的15对大麦5HS染色体EST 标记中,共显性SNP 标记K05820共检测出109株抗性亲本纯合体、115株感病亲本纯合体和233株杂合体。利用F2代表型分离结果对标记K05820进行验证分析发现,该标记与‘Pompadour’5HS染色体成株叶锈病抗性高度相关(r=0.82)。同时,新的定位分析表明,K05820 与成株叶锈病抗性基因间的遗传距离为7 cM。这说明共显性EST SNP标记K05820 可以运用于大麦5HS染色体成株叶锈病抗性基因后代杂合体及纯合体的筛选,能够应用于田间抗病材料的初步筛选。

      The research was conducted to find out a useful marker from barley 5HS EST to analyze the F2 population derived from an adult plant leaf rust resistant variety ‘Pompadour’. For mining single nucleotide polymorphism (SNP) markers, single strand conformation polymorphism (SSCP) technique was used to distinguish heterozygous and homozygous in 457 F2 population derived from ‘WABAR2482×Pompadour’. F2 phenotype data demonstrated that 328 plants were found in resistant group and 129 plants were included in the susceptible group. The new gene was postulated as a major dominant gene controlling the adult plant resistance on barley 5HS by a segregation ration 3∶1. Besides, the co dominant EST SNP marker K08520 tested 109 resistant homozygous, 115 susceptible homozygous and 233 heterozygous in 457 F2 population. Association analysis demonstrated that correlation coefficient between the marker K08520 and the adult plant resistance was 0.82, which shows that EST SNP K08520 could be used in the molecular assisted selection (MAS) for the adult plant resistance (APR) to barley leaf rust in Pompadour. In addition, the segregation data displayed that K08520 was mapped 7cM to the new APR gene. The new developed EST SNP marker K08520 is a useful molecular marker for analyzing heterozygous, homozygous and MAS for the adult plant resistance in ‘WABAR2482×Pompadour’ F2 population.
