      为进一步了解硫氧还蛋白过量表达促进大麦种子萌发的机理,以硫氧还蛋白S基因(Trxs)过量表达的转基因大麦纯合株系(LSY 11 1 1)及其非转基因对照(CK)为试验材料,研究了Trxs过量表达对大麦成熟种子中硫氧还蛋白h(Trxh)催化活性、巯基含量、激素含量、蛋白组分及吸水速率的影响。结果显示:① 转基因大麦成熟种子中Trxh催化活性显著高于CK(P<0.01);转基因大麦种子的还原状态增强,LSY 11 1 1种子中巯基含量显著高于CK(P<0.01);转基因大麦成熟种子的激素含量增加,其中LSY 11 1 1的赤霉素(GA)与生长素(IAA)含量显著高于CK(P<0.05)。② 转基因大麦成熟种子中醇溶蛋白与CK明显不同,高分子量蛋白条带数与丰度高于CK。③ 转基因大麦的吸水速率比CK快,在萌发10、30和50 h时,LSY 11 1 1的含水量分别比CK增加了21.87%、15.38%和11.11%,差异显著(P<0.05)。上述结果表明,Trxs过量表达可以通过改变转基因大麦成熟种子中巯基含量、激素含量和醇溶蛋白组分等方式影响大麦种子萌发过程中的C、N转移效率,从而促进萌发进程。
英文摘要:
      To clarify the function of overexpressing thioredoxin S(Trxs) gene in transgeneic barley in promoteing germination process, the barley variety, LSY(CK)and its transgenic line with overexpressing Trxs (LSY 11 1 1) were employed to measure the changes of physiological, bio chemical characteristics and protein ingredients in mature barley seeds. The Trx h activity of transgenic barley was significantly higher than CK(P<0.01)in mature seeds. the Trx h activity of LSY 11 1 1 was 62.97% higher than CK at the 41st minute after reaction. The reducing state of transgenic barley seeds was increased, as reflected by the significantly higher contents of sulphydryl(P<0.01). The phytohormone content of transgenic barley was higher than CK, in which the gibberellic acid (GA) and indoleacetic acid (IAA) contents were significantly higher than CK(P<0.05). (ii) The state of hordein in seeds was distinctly different between LSY 11 1 1 and CK, especially the bands and abundance of high molecular weight protein was more plentiful than CK. (iii) In comparison with CK, the rate of water absorption in the transgenic barley seeds was significantly greater(P<0.05). The water content of transgenic seeds is 21.87%, 15.38% and 11.11% higher than CK at 10 h, 30 h and 50 h after germination, respectively. All the results indicated that overexpressing Trxs efficiently altered sulphydryl content, phytohormone content and hordein state in seeds of transgenic barley, which laid an important basis for promoteing germination process of barley seeds through increasing transfer efficiency of carbon (C) and nitrogen (N).
