为了克隆节节麦中的HMW GS基因,利用十二烷基硫酸钠聚丙烯酰胺凝胶电泳(SDS PAGE)分析了节节麦的HMW GS,发现1种编码序列未知的y 型亚基,即Dy8.1t亚基。通过对目的片段的AS PCR扩增、克隆、测序和氨基酸序列推导,发现这种未知序列具有典型HMW GS的序列结构特征。通过与已知亚基序列比较发现,Dy8.1t与Dy10t、Dy10.4t、Dy10.5t和Dy12t在N 端序列有一个氨基酸差别(R G)。与已知氨基酸序列的HMW GS多序列比对和系统进化关系分析,证实Dy8.1t亚基是D基因组编码的高分子量谷蛋白y 型亚基家族的新成员。 |

Triticum tauschii( 2n=2x=14, DD), as the D genome donor of common wheat, contains a wide range of high molecular weight glutenin subunit (HMW GS) alleles, and has more genetic variation than the common wheat genome. We used sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS PAGE) to analyze the HMW GS of Aegilops tauschii, and one new y type subunit, designated Dy8.1t was found. Through AS PCR amplification and DNA sequencing, Dy8.1t was cloned, with a typical sequence of HMW GS. Compared with the known subunits Dy10t, Dy10.4t, Dy10.5t and Dy12t, Dy8.1t had an amino acid difference (R G) in the N terminal sequence. Based on multiple sequence alignment and phylogenetic relationship analysis, Dy8.1t as a new member of HMW glutenin y type subunits were confirmed. |
