In order to study the effect of barley stripe pathogen toxin on barley seed germination,seedling growth and defensive enzymes activity,20 barley varieties were treated with the barley stripe pathogen toxin. The inhibition rate of seed germination,radicle and germ growth of the toxin after 120 h were measured. The activities of superoxide dismutase(SOD),catalase(CAT),peroxidase(POD) and the contents of free proline(Pro) and malondialdehyde(MDA) of variety with strong disease resistance were measured at 48 h,72 h,120 h and 168 h after germination.The results showed that 20 barley varieties were divided into three groups according to the germination inhibition rate,i.e. high resistance type(5%),medium resistance type(60%) and high sensitivity type(35%). The differences of germination indices among the three types were significant(P<0.05). As time went on,the activities of SOD and CAT in the seedlings of Ganpi 2,a strong resistant cultivar treated with toxin solution,decreased first and then increased,and the activities of SOD and CAT were the lowest at 72 h; the activities of POD and content of Pro showed an upward trend; the content of MDA increased first and then decreased. The activities of SOD,POD,CAT and content of Pro and MDA in Ganpi 2 treated with toxin solution were higher than those of control. In conclusion,the toxin of barley stripe pathogen can inhibit the growth of radicle and germ of barley seeds to a certain extent; barley plants can improve their resistance by enhancing the activity of defensive enzymes; the varieties with resistance can be screened out by using the toxin to treat seeds. The results of this study can provide a basis for the study of resistance mechanism and breeding of resistant varieties. |
