      菲利普孢囊线虫 (Heterodera filipjevi) 是严重危害我国黄淮海地区小麦生产的重要病原物之一。为揭示该线虫侵染后合胞体建立早期病原与宿主细胞的互作机制,以H.filipjevi易感材料温麦19和抗性材料抗线2号为研究对象,应用real time PCR分析了线虫侵染后不同时段10个可能与防卫相关的UniGenes相对表达量的变化。结果表明,与细胞结构相关的基因UniGene AK335102(GenBank ID,下同)在抗线2号中的表达量高于温麦19,而另一个与细胞结构相关的UniGene DR737925在温麦19中的表达量高于抗线2号。与应激相关的一个UniGene JV948284在侵染后各时段,在温麦19中的表达量均远高于抗线2号。与抗性相关的UniGene JP233598在接种线虫3 d以后,在抗线2号中的表达量明显高于温麦19;UniGene JP852719(Ascorbate peroxidase) 基因在抗、感材料中的表达量无明显差异;UniGene(CV767657) TIFY 3A-like protein在温麦19中的表达量随接种后时间延长而降低,而在抗线2号中的表达量仅在接种后3、9 d时的表达量相对较高。信号转导相关UniGene CD87797与WRKY转录因子家族UniGene EU665453,在温麦19中的表达量随接种后时间延长而逐渐增加,但在抗线2号中则逐渐降低。MYB 转录因子UniGene JF951950的表达量在温麦19中接种早期较高,而在抗线2号中接种后期较高;另一MYB 转录因子UniGene AK330737(similarity to Os06g0258000) 在抗线2号中的表达量明显高于温麦19。综上所述,在菲利普孢囊线虫侵染后的早期,10个可能与防卫相关的基因表达模式在抗性和易感材料中存在不同程度差异,此结果对揭示小麦对菲利普孢囊线虫抗、感的分子机制有一定启示。
英文摘要:
      Heterodera filipjevi is one of the main pests that seriously endanger wheat production in Huanghuaihai region of China. In order to reveal the interaction mechanism between pathogen and host cells in the early stage of syncytium formation after infection by Heterodera filipjevi,this study was conducted to analyze the expression patterns of putative 10 defense-related genes at different incubation time using real-time PCR with cultivars susceptible and resistant to Heterodera filipjevi. The results showed that the UniGene AK335102(GenBank ID,the same below),one of the cell-structure related genes,was highly expressed in resistant cultivar Kangxian 2 than in susceptible cultivar Wenmai 19,while UniGene DR737925,another gene related to cell structure,was highly expressed in Wenmai 19 than in Kangxian 2.The expression of stress-related UniGene JV948284,an unchracterized protein,in Wenmai 19 was much higher than that in Kangxian 2 at different time after infection. For genes related to resistance,the expression of UniGene,ie,JP233598,in Kangxian 2 was significantly higher than that in Wenmai 19 after 3-day inoculation. There was no significant difference in the expression of JP852719 between resistant and susceptible cultivars. In addition,the expression of Ascorbate peroxidase(WAPX1),ie UniGene CV767657,in Wenmai 19 was decreased with the progress of inoculation time,while that in Kangxian 2 was relatively high only at 3 and 9 days after inoculation. The expression of signal transduction related UniGene CD877975(Tubulin/FtsZ family protein) and WRKY transcription factor UniGene EU665453 was increased dramatically in Wenmai 19,but was decreased significantly in Kangxian 2 with the progress of inoculation time. The expression of MYB transcription factor UniGene JF951950 was higher at the early stage of inoculation in Wenmai 19,and higher at the late stage of inoculation in Kangxian 2. The expression of another MYB transcription factor UniGene AK330737,in Kangxian 2 was significantly higher than that in Wenmai 19. It can therefore be concluded the expression patterns of 10 putative defense-related genes in the early post-infection period of Heterodera filipjevi differ between resistant and susceptible cultivars,which can explain the molecular mechanism underlying the resistance and susceptibility of wheat to Heterodera filipjevi.
